When booking a specific hotel directly on hotwire,  most of the time extra guest charge is added for the two children, although room rate includes two queen beds. What to expect when booking the deal above,  can the hotel still add an extra guest fee for our two kids once we arrived?

They shouldn't if you include 2adults/2children in your search.

If there was an issue i think you would win this argument with the hotel and/or HOTWIRE... but if you want to avoid the situation entirely just have one parent go to the desk at check-in while the other two wait someplace else in the lobby or start to wonder around the hotel.
