There are a LOT of factors affecting whether you can get an upgrade or not... occupancy levels during your stay, what is available at time of check-in, mood of the front desk clerk, hotel policy, etc, etc.

It's Vegas, so you can certainly try if you'd like, but my advice is to always research ahead of time to find out exactly what types of rooms are offered at the hotel and what type of upgrade it is that you want... too often everyone wants an upgrade, and settles for the 'Strip View' which isn't really what they originally had in mind.
