If I do my search in HOTWIRE with 2 adults and 2 children, am I guaranteed a 2 bed room? What about PRICELINE?

PRICELINE only guarantees one bed/double occupancy. One can request two beds with the hotel, but the hotel can definitely say no or charge more.

With HOTWIRE, if you search for more than 2 people, you are ensured of some form of bedding (usually two beds, but can be 1 + sofa or other combination) for the party. If you search for 2 or less, you're likely looking at one bed. Again, you can request 2, but the hotel can definitely say no.
